I am planning a driving tour of Tuscany for 2 full days and need recommendations for best hill towns with dining an winery recommendations

Have you already decided on your home base in Tuscany? If not, my suggestion is you base near the wonderful city of Siena, may in one scenic place in the countryside. That location will be great to explore Chianti and the Val d’Orcia, the 2 most important wine regions of Tuscany. Both areas are also among the most sensational of Tuscany in relation to their scenic landscape. The best hill towns with great food and great wineries are right in Chianti and Val d’Orcia, so you can’t go wrong! 
In Chianti, I recommend Castellina in Chianti, Greve in Chianti, and Gaiole in Chianti. You eat well practically everywhere. In the same hill town you’re going to find wine bars and wine shops where to taste wine. A great places to eat in Chianti is the Osteria di Passignano, and if you love the famous Florentine steak, do go for the Officina delle Bistecca in Panzano in Chianti. 
In Val d’Orcia don’t miss Montalcino, Pienza, and Montepulciano. This area produces some of the best wines of Italy, like the Brunello di Montalcino. Pienza is the place for great dinners, the cheese called pecorino di Pienza is used to make unforgettable dishes; Osteria Bacchus is to be recommended!

We will be based in an apt close to Poppiano then will drive to Amalfi coast. Will do more research. Thanks!

The area surrounding the Castle of Poppiano is beautiful and has a central location in Tuscany. You can in fact reach Chianti in less than 40 minutes, so do go for Chianti on one day. The Castello di Verrazzano is another beautiful place in Chianti where to go for wine tastings. You may also include San Gimignano on your first day; this beautiful medieval hill town, famous for its scenic towers, also produces a fine white wine called Vernaccia di San Gimignano.
Montalcino and Montepulciano are just at 1 hour and half driving, so also the Val d’Orcia is doable! In both villages are really many restaurants and wine bars and you eat well practically everywhere.
